{"id":24856,"date":"2020-07-10T17:56:00","date_gmt":"2020-07-10T14:56:00","guid":{"rendered":"https:\/\/en.buradabiliyorum.com\/researchers-solve-a-50-year-old-enzyme-mystery\/"},"modified":"2020-07-10T17:56:00","modified_gmt":"2020-07-10T14:56:00","slug":"researchers-solve-a-50-year-old-enzyme-mystery","status":"publish","type":"post","link":"https:\/\/buradabiliyorum.com\/en\/researchers-solve-a-50-year-old-enzyme-mystery\/","title":{"rendered":"#Researchers solve a 50-year-old enzyme mystery"},"content":{"rendered":"<p>&#8220;<strong>#Researchers solve a 50-year-old enzyme mystery<\/strong>&#8221;<\/p>\n<div>\n<div>\n<div data-src=\"https:\/\/scx2.b-cdn.net\/gfx\/news\/2020\/uqresearcher.jpg\" data-sub-html=\"Credit: The University of Queensland\" data-thumb=\"https:\/\/scx1.b-cdn.net\/csz\/news\/tmb\/2020\/uqresearcher.jpg\">\n<figure><img loading=\"lazy\" decoding=\"async\" alt=\"UQ researchers solve a 50-year-old enzyme mystery\" height=\"480\" src=\"https:\/\/scx1.b-cdn.net\/csz\/news\/800\/2020\/uqresearcher.jpg\" title=\"Credit: The University of Queensland\" width=\"800\"><\/img><figcaption>\n                Credit: The University of Queensland<br \/>\n            <\/figcaption><\/figure>\n<\/div>\n<\/div>\n<p>Advanced herbicides and treatments for infection may result from the unraveling of a 50-year-old mystery by University of Queensland researchers.<\/p>\n<section>\n      <\/section>\n<p>The research team, led by UQ&#8217;s Professor Luke Guddat, revealed the complete three-dimensional structure of an enzyme, providing the first step in the biosynthesis of three essential amino acids\u2014leucine, valine and isoleucine.<\/p>\n<p>&#8220;This is a major scientific advance, which has been pursued globally by chemists for half a century,&#8221; Professor Guddat said.<br \/>\n&#8220;This information provides new insights into an important enzyme\u2014acetohydroxyacid synthase\u2014a target for more than 50 commercial herbicides.<br \/>\n&#8220;It&#8217;s also a potential target for new drugs to treat infections such as tuberculosis and invasive Candida infections.&#8221;<br \/>\nUsing advanced techniques such as cryo-electron microscopy and X-ray crystallography, the team deciphered the structure of the plant and fungal versions of the enzyme.<br \/>\n&#8220;We identified how this highly complex structure is assembled, which is the highly unusual shape of a Maltese Cross,&#8221; Professor Guddat said.<br \/>\n&#8220;Coincidently, the Maltese Cross also features as a part of UQ&#8217;s logo.&#8221;<br \/>\nProfessor Guddat said the discovery could have big implications for global agriculture.<br \/>\n&#8220;Sulfometuron is a herbicide that targets this enzyme, and was widely used in the 1990s for wheat crop protection throughout Australia,&#8221; he said.<br \/>\n&#8220;But today it is completely ineffective due to the development of resistance.<br \/>\n&#8220;With this new insight, we will be able to make changes to existing herbicides, restoring options for future herbicide <a href=\"https:\/\/buradabiliyorum.com\/en\/category\/download-scripts-themes-apps\/\" data-internallinksmanager029f6b8e52c=\"9\" title=\"Download Scripts &amp; Themes &amp; Apps\" target=\"_blank\" rel=\"noopener\">app<\/a>lication.&#8221;<br \/>\nProfessor Guddat said the enzyme was only found in plants and microbes, not in humans.<br \/>\n&#8220;For this reason, the herbicides and drugs that it targets are likely to be safe and non-toxic to all mammals,&#8221; he said.<br \/>\n&#8220;And another surprising finding of the research was the role that the molecule known as ATP plays in the regulation of the enzyme.<br \/>\n&#8220;Normally ATP plays a role in providing energy to all living cells,&#8221; Professor Guddat said.<br \/>\n&#8220;However, here it is acting like a piece of glue to hold the structure together.&#8221;<br \/>\n&#8220;They&#8217;re fascinating findings for us, and we&#8217;re excited for new opportunities for targeted design of next-gen herbicides and antimicrobial agents.&#8221;<\/p>\n<hr>\n<\/hr>\n<hr>\n<\/hr>\n<div>\n<p><strong>More information:<\/strong><br \/>\n                                                Thierry Lonhienne et al, Structures of fungal and plant acetohydroxyacid synthases, <i>Nature<\/i> (2020).
